چکیده
Crystallization is a key step in macromolecular structure determination by crystallography. While a robust theoretical treatment of the process is available, due to the complexity of the system, the experimental process is still largely one of trial and error. In this article, efforts in the field are discussed together with a theoretical underpinning using a solubility phase diagram. Prior knowledge has been used to develop tools that computationally predict the crystallization outcome and define mutational approaches that enhance the likelihood of crystallization. For the most part these tools are based on binary outcomes (crystal or no crystal), and the full information contained in an assembly of crystallization screening experiments is lost. The potential of this additional information is illustrated by examples where new biological knowledge can be obtained and where a target can be sub-categorized to predict which class of reagents provides the crystallization driving force. Computational analysis of crystallization requires complete and correctly formatted data. While massive crystallization screening efforts are under way, the data available from many of these studies are sparse. The potential for this data and the steps needed to realize this potential are discussed.

The failure of multi-step experimental procedures to yield diffraction-quality crystals is a major bottleneck in protein structure determination. Accordingly, several bioinformatics methods have been successfully developed and employed to select crystallizable proteins. Unfortunately, the majority of existing in silico methods only allow the prediction of crystallization propensity, seldom enab...
متن کاملA computational model for processing of semi-crystalline polymers: The effects of flow-induced crystallization
A computational model for the combined processes of quiescent and flowinduced crystallization of polymers is presented. This modelling should provide the necessary input data, in terms of the structure distribution in a product, for the prediction of mechanical properties and shapeand dimensional-stability. Rather then the shear rate as the driving force, a viscoelastic approach is proposed, wh...
متن کاملUsing Machine Learning to Analyze Biological Macromolecular Crystallization Data
The crystallization of a new macromolecule is still very much a trial and error process. In an e ort to uncover useful trends in the crystallization of new macromolecules, Samudzi, Fivash and Rosenberg[12] performed a cluster analysis on the Biological Macromolecule Crystallization Database(BMCD)[7]. The crystallization parameters that were studied in order to di erentiate among the experiments...
متن کاملA computationally efficient inorganic atmospheric aerosol phase equilibrium model (UHAERO)
A variety of thermodynamic models have been developed to predict inorganic gasaerosol equilibrium. To achieve computational efficiency a number of the models rely on a priori specification of the phases present in certain relative humidity regimes. Presented here is a new computational model, named UHAERO, that is both efficient and 5 rigorously computes phase behavior without any a priori spec...
متن کاملUsing Computational Modeling to Control and Optimize the Crystallization Process
Crystallization processes are used across the chemical, petrochemical and pharmaceutical industries. In the chemical industry, the potash and caustic industries use crystallization as a means to produce bulk chemicals and also as a separation technology for purification. The fine chemical and specialty chemical industries use crystallization processes to produce various high-value organic compo...
